Description

Description: This assay employs a two-site sandwich ELISA to quantitate CLDN11 in samples. An antibody specific for CLDN11 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CLDN11 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CLDN11 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CLDN11 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Claudin-11 belongs to the claudin family of tight junction associated proteins and is a major component of central nervous system myelin that is necessary for normal CNS function. There is growing evidence that the protein determines the permeability between layers of myelin sheaths via focal adhesion and, with its expression highly regulated during development, may play an important role in cellular proliferation and migration. In addition, the protein is a candidate autoantigen in the development of autoimmune demyelinating disease.Its nucleotide sequence shares little homology with previously reported genes, but the predicted amino acid sequence and structure of the gene product suggest that the protein, which they symbolized Osp, is the central nervous system (CNS) homolog of peripheral myelin protein-22
